you either make it or easier way is just to open the original then copypaste what you made over it and save with the alpha in there.
edit:
now that i looked at the pic seems it's a weapon glow... and i'm guessing you just painted the whole dds white and then saved. the file needs some black in there (to get some non visible stuff) otherwise you'll get squares.
