Well it certainly depends where the Checkbox is located. If it's on the self-made GUI then GUICtrlSetState is the way to go (although you could assign an Accelerator for that Ctrl to make it easier) - If the Checkbox is located on a foreign GUI then I'd recommend to use ControlCommand / ControlClick.
